{"id":1160,"date":"2018-10-04T11:35:31","date_gmt":"2018-10-04T11:35:31","guid":{"rendered":"http:\/\/priorclavena.com\/?p=1160"},"modified":"2021-12-21T04:06:49","modified_gmt":"2021-12-21T04:06:49","slug":"research-lab-autoclave-best-practices-the-liquid-cycle-wont-save-you","status":"publish","type":"post","link":"https:\/\/www.priorclave.com\/en-us\/2018\/10\/04\/research-lab-autoclave-best-practices-the-liquid-cycle-wont-save-you\/","title":{"rendered":"Research Lab Autoclave Best Practices: The Liquid Cycle Won&#8217;t Save You"},"content":{"rendered":"<p class=\"p3\">Every modern research lab autoclave has a &#8220;liquid cycle&#8221;\u2014a cycle <i>just<\/i> for <a href=\"https:\/\/www.priorclave.com\/en-us\/liquids-sterilizers-autoclaves\/\">processing liquids<\/a>, growth media, and so on.<span class=\"Apple-converted-space\">\u00a0 <\/span>Nonetheless, more often than not, if you fill a bunch of 1L bottles up to 900mL and process them on the &#8220;liquid cycle&#8221; they&#8217;re going to come out like this:<\/p>\n<p class=\"p3\">The glass isn&#8217;t cracked; where did the liquid go?<span class=\"Apple-converted-space\">\u00a0 <\/span>Some evaporated, but most of it boiled up out of the flasks and fouled the inside of the autoclave.<\/p>\n<p class=\"p3\">What&#8217;s the solution?<span class=\"Apple-converted-space\">\u00a0 <\/span>Just tighten the lids so nothing escapes?<\/p>\n<p class=\"p3\"><i>NO!<\/i> Never, ever autoclave a sealed container; <a href=\"http:\/\/www.priorclave.com\/en-us\/2018\/09\/20\/research-lab-autoclave-safety-sealed-bottles-are-bombs\/\">it becomes a bomb<\/a>.<\/p>\n<p class=\"p3\">There are two safe, effective ways to address boil over.<span class=\"Apple-converted-space\">\u00a0 <\/span>They&#8217;re cheap and easy:<\/p>\n<ul class=\"ul1\">\n<li class=\"li3\">Use bigger bottles<\/li>\n<li class=\"li3\">Chose the right autoclave cycle<\/li>\n<\/ul>\n<h2 class=\"p3\">Running Research Lab Autoclave Liquid Cycles in the Real World<\/h2>\n<p class=\"p3\">&#8220;<a href=\"http:\/\/www.priorclave.com\/en-us\/2018\/09\/06\/top-tips-for-autoclaving-pipette-tips\/\">Dry cycles<\/a>&#8221; tend to either pull a vacuum on the chamber, or permit one to naturally form.<span class=\"Apple-converted-space\">\u00a0 <\/span>Either way, lowering the chamber pressure lowers the boiling point in the chamber (due to the aptly named Boyle&#8217;s Law).<span class=\"Apple-converted-space\">\u00a0 <\/span>Liquid more readily vaporizes at lower pressure, and you end up with dry (or, at least, <i>dryer<\/i>) loads.<span class=\"Apple-converted-space\">\u00a0 <\/span>A &#8220;liquid cycle&#8221; never pulls a vacuum and makes a point of venting more slowly, so a vacuum will not form.<span class=\"Apple-converted-space\">\u00a0 <\/span>Ideally, this means little evaporation and no boil up.<\/p>\n<p class=\"p3\"><i>&#8220;Ideally.&#8221;<\/i><\/p>\n<p class=\"p3\">Sadly, this is the real world, not the ideal whiteboard of a physics classroom.<span class=\"Apple-converted-space\">\u00a0 <\/span>Pressure and temperature vary throughout the cycle, especially as the cycle ends.<span class=\"Apple-converted-space\">\u00a0 <\/span>You can expect some evaporation (most labs assume 10% to 20%) and expansion as gases precipitate out of the fluid.<span class=\"Apple-converted-space\">\u00a0 <\/span>Frothing is almost inevitable, and is especially the case with broths, growth media, and agars, and most pronounced with LB agar.<\/p>\n<p class=\"p3\">A good way smooth these temperature and pressure fluctuations is to take your bottles of liquid, set them in a tray or tub, and pour some water into the bottom of the tub. Water absorbs and releases heat more slowly than air or steam; having your bottles rest in a water bath acts as a buffer and smooths out the temperature and pressure
